ABSTRACT

ObjectiveProteomics was used to investigate the protein differences between porcine cardiac blood(PCB) and porcine blood(PB) from Menghe medical school and to compare the effects of both on the microglial inflammation of Salviae Miltiorrhizae Radix et Rhizoma(DS). MethodNanoliquid chromatography-quadrupole orbitrap mass spectrometry(nLC-MS/MS) and bioinformatics were utilized to compare the proteomic differences of PCB and PB in simulated gastrointestinal digestion. Furthermore, Western blot was used to verify the contents of some shared proteins and differential proteins identified in PCB and PB. In addition, BV2 neuroinflammation model constructed by corticosterone(CORT) and lipopolysaccharide(LPS) was applied to detect the intervention effects of PCB and PB on the levels of tumor necrosis factor(TNF)-α and interleukin(IL)-6 in BV2 inflammatory cells of DS. ResultA total of 69 common proteins and 68 differential proteins were identified in PCB and PB, among which the common proteins included transferrin(Tf) with brain-targeting effect, and the differential proteins in the two were 41 and 27, respectively. Western blot validation showed that the difference in the content of the same protein Tf between PCB and PB was not statistically significant, while the difference in the contents of the specific proteins of creatine kinase M and heart-type fatty acid binding protein(H-FABP) were statistically significant(P<0.05). Moreover, in vitro experimental studies revealed that compared with the same concentration of DS group, in addition to the 100 mg·L-1 PB-DS group, PCB-DS and PB-DS groups could significantly inhibit the levels of TNF-α and IL-6 in BV2 inflammatory cells(P<0.05, P<0.01), and PCB-DS group had more significant anti-inflammatory effect than PB-DS group with the same concentration(P<0.05, P<0.01). ConclusionBoth of PCB and PB can enhance the inhibitory effect of DS on the release of inflammatory factors, thus playing a neuroprotective role, and PCB promotes DS inhibition more significantly, which may be due to the existence of the two involved in energy metabolism-related differential proteins, which can lay a foundation for revealing the scientific connotation of the processing of PCB-DS and PB-DS.

ABSTRACT

Objective: To compare the chemical components in raw products and characteristic processed products with porcine cardiac blood of Salviae Miltiorrhizae Radix et Rhizoma from Menghe medical school.Method: The ultra performance liquid chromatography-quadrupole-time of flight mass spectrometry (UPLC-Q-TOF/MS) was performed on an ACQUITY UPLC® C18 column (2.1 mm×100 mm,1.7 μm),mobile phase was 0.1% formic acid aqueous solution (A)-0.1% formic acid acetonitrile solution (B) for gradient elution (0-1 min,8% B,1-1.5 min,8%-20% B,1.5-4 min,20% B,4-5 min,20%-60% B,5-9 min,60%-70% B,9-10 min,70%-95% B,10-13 min,95% B).The column temperature was 40℃ and the flow rate was 0.3 mL·min-1.Q-TOF/MS with electrospray ionization (ESI) and scanning range of m/z 50-1 200 were applied for analysis under positive and negative ion mode,respectively.All ionic peaks were assigned by comparison of reference substances,mass spectra data,database matching and literature reference,changes of chemical components in Salviae Miltiorrhizae Radix et Rhizoma were investigated by comparing number and area of ionic peaks before and after processing.Result: A total of 59 components were identified from raw products and characteristic processed products,and peak areas of 25 components showed obvious change.However,there were no new compound was found in characteristic processed products.After being processed with porcine cardiac blood,the contents of water-soluble ingredients (protocatechuic aldehyde,salvianolic acid C,F,G),fat-soluble ingredients (tanshinaldehyde,tanshindiol A,tanshinone Ⅰ) and amino acid (L-phenylalanine) in Salviae Miltiorrhizae Radix et Rhizoma were significantly increased.Conclusion: Changes of contents of chemical components in raw products and processed products of Salviae Miltiorrhizae Radix et Rhizoma are remarkable,part of water-soluble ingredients,fat-soluble ingredients and amino acids have quantitative change,which may be related to promoting treatment of Salviae Miltiorrhizae Radix et Rhizoma on cerebral ischemia after being processed with porcine cardiac blood.

ABSTRACT

The porcine cardiac blood processed Salvia miltiorrhiza (PCB Danshen) is the characteristic processing of Menghe medical school and has been inherited for hundreds of years, commonly used in the treatment of brain ischemia-induced agitation, palpitation and phlegm confusing heart. Ancient and modern physicians believe that porcine cardiac blood is a guiding for heart nourishing drugs, which could enhance the effects of Salvia miltiorrhiza by nourishing and soothing the nerves. However, the material basis and processing mechanism of PCB Danshen are still unclear. This paper investigated the historical evolution and modern research of PCB Danshen, including the clinical application, the intention of clinic processing, the processing technology and recent research of PCB Danshen. In addition, the major problems and significance in research and development of PCB Danshen were further thought and prospected, hoping to provide basic data for material basis and processing mechanism of PCB Danshen, and provide effective support for inheriting and carrying forward the characteristic processing technology of Menghe medical school.

ABSTRACT

[Objective]By analyzing the treatment of stomachache of the surnames FEI and MA from Menghe scholarly sect, exploring the experiences in treatment and formula regularity. [Method]The thesis gives standard of medicine names and processing technology based on Traditional Chinese Pharmacology;calculates the medication frequency and classifies medicines by frequency analysis.[Results]There are totally 40 medical cases included in the study, statistics analysis shows that the drug is mainly Qi regulating drugs,drugs for eliminating dampness, drugs of promoting blood circulation and hematinics drugs.Mr FEI took 13.22 drugs on average,high-frequency medicines include Angelica,Tribulus terrestris,Tangerine Peel,Green Tangerine Peel, Radix aucklandiae.Mr MA took 10.72 drugs on average,high-frequency medicines include Salvia miltiorrhiza,Poria cocos,Angelica,Radix Curcumae,Pinellia. [Conclusions]The surnames FEI and MA from Menghe scholar sect paid attention to the method of regulating qi-flowing for harmonizing stomach,the method of resolving phlegm and removing blood stasis,the method of pacifying the stomach through the treatment of the liver.The idea of prescribing is moderate, lightly and well-proportioned.The nuances state their personal characteristics.It's worth our reference in clinical treatment.

ABSTRACT

[Objective]To explore the clinical experience of professor SHAN Zhaowei treatment of chronic atrophic gastritis(CAG). [Method] To analyze SHAN Zhaowei professor from the perspective of four different pathogenesis of syndrome differentiation of CAG. To sum up professor SHAN Zhaowei 's clinical experience in the treatment of chronic atrophic gastritis, inheritance of Meng He YiPai essence. [Result] Professor SHAN Zhaowei treats CAG in qi, deficiency, blood stasis and toxin from four different pathogenesis to syndrome differentiation of traditional Chinese medicine treatment of CAG, mediation or insufficiency of qi activity, health, and remove stasis coating, detoxification cancer, the first emphasis on TCM four diagnosis, tongue mirror each other, differentiation is exquisite, precision, protecting stomach qi, along with the differentiation to add and subtract, insipid, but with magic effect. [Conclusion] The unique pathogenesis differentiation method in the treatment of CAG and the clinical experience for CAG of Professor SHAN Zhaowei is worth learning and promotion.

ABSTRACT

Objective]To investigate academic thoughts of Prof. Yan Dexin in spleen and stomach syndrome treatment.[Methods]The paper summarized the academic origin, academic thoughts as well as practical treatment cases of Prof. Yan Dexin ’s spleen and stomach syndrome treatment theory. [Results]The main points of Prof. Yan’s academic thought on spleen and stomach therapy has succeeded and developed the therapy of Menghe physician, which includes an emphasis on spleen Qi rise and stomach Yin drop, Qi of spleen and stomach regulation, special attention on the function of the stomach-yang and spleen-yin, an advocation ofspleen leading four organs, and therapy design from spleen with an ingenious use of atractylodes rhizome, Atractylodes and Zhizhu pill. [Conclusion] Professor Yan Dexin developed his unique theoretical system in spleen-stomach diseases therapy on basis of the experiences of spleen and stomach disease treatment from ancient physicians, which is instructive for later physicians.
